Synthesis Analysis
The total synthesis of GLA has been achieved through a series of complex organic reactions, highlighting its intricate molecular architecture. A notable synthesis approach involves Mn(OAc)3-mediated radical cyclization of alkynyl ketones, paving a new avenue for the synthesis of highly oxidized diterpenoids, including GLA. This method features a highly enantioselective conjugate addition/acylation cascade reaction, a Yamamoto aldol reaction, and an intramolecular Diels-Alder reaction to assemble the A/B ring system, demonstrating the synthetic challenge and complexity of GLA .
