Description of Dapivirine CAS NO:244767-67-7 

Dapivirine CAS NO:244767-67-7  is an antiviral drug of the non-nucleoside reverse transcriptase inhibitor class. It was developed for the potential treatment and prevention of HIV infection in humans by acting as a competitive inhibitor of the viral enzyme reverse transcriptase. This drug has been shown to have anti-HIV activity in vivo, and was found to be safe even at high doses in animals. Dapivirine is also active against herpes simplex virus type 2 (HSV-2). The molecule was originally synthesized in 1979, but it took until 2006 before it was approved for human clinical trials. In vitro studies show that dapivirine inhibits the transcriptional regulation of HIV-1 LTR and prevents replication of HIV-1 RNA.

Pharmacokinetic studies of Dapivirine vaginal microbicide gels have provided critical insights into the drug's safety and delivery mechanisms. These studies have shown that Dapivirine can be delivered to the lower genital tract at concentrations significantly higher than its in vitro inhibitory concentrations, suggesting effective protection against HIV infection with minimal systemic exposure


Future Directions
The World Health Organization has recommended that the dapivirine vaginal ring may be offered as an additional prevention choice for women at substantial risk of HIV infection as part of combination prevention approaches . Research is also underway to develop a vaginal ring that includes both contraception and HIV prevention
