The MOF material used is PCN-333 (Al), which has two types of mesopores with diameters of 4.2 nanometers and 5.5 nanometers respectively.
PCN-333 was selected for three reasons: high enzyme loading capacity, easy modification with fluorophores (for easy tracking in subsequent biological experiments), and stable structure in the cell environment. Since tyrosinase is a relatively large enzyme with a molecular weight of 43 kDa and a size of about 5.5 × 5.5 × 5.6 cubic nanometers, only the pores of PCN-333 with a diameter of 5.5 nanometers can accommodate the enzyme.
After mixing the PCN-333 nanoparticles (NPCN-333) with tyrosinase in the solution for 20 minutes, the enzyme loading reached 0.80 g/g, close to the theoretical loading of 1.08 g/g. From the pore size distribution, the 5.5 nanometer pores in the enzyme-loaded MOF-TYR@NPCN-333 are occupied by the enzyme.
